Born on June 20, 1931, in Calumet, Michigan, Tolkan faced early challenges when his parents divorced. At age 14, he cycled through Chicago and settled in Tucson, Arizona, graduating from Amphitheater High School in 1949. He briefly served in the Navy during the Korean War and attended colleges including Coe College and the University of Iowa. With just $75 in his pocket, he headed to New York City, worked on the docks, and later trained at The Actors Studio under Stella Adler and Lee Strasberg.

Tolkan dedicated 25 years to New York theater before transitioning to film. He collaborated frequently with director Sidney Lumet in projects like Serpico (1973), Prince of the City (1981), and Family Business (1989). In 1983, he relocated to California and Canada for WarGames.
His standout roles include the stern Mr. Strickland across the Back to the Future films (1985, 1989, 1990), evolving from the principal to his grandfather in the third installment. He also commanded attention as Stinger opposite Tom Cruise in Top Gun (1986).
